Dottie Peoples was born on 12 August 1950 in Dayton, Ohio, as one of ten children to Althea and Robert Milton. As a child, she spent her summers with her grandmother in Birmingham, Alabama. During these visits, she experienced long Sunday worship services filled with traditional gospel music and discovered the recordings of famous gospel singer Mahalia Jackson. Inspired by Jackson, Dottie once told her grandmother that she wanted to follow in her footsteps and become a gospel singer herself.

After finishing high school, Dottie Peoples sang with gospel singer Dorothy Norwood and later toured with Shirley Caesar. She also performed with a jazz group led by Groove Holmes, though she kept this a secret from her mother. After getting married, she moved to Atlanta, where she became the music director at Salem Baptist Church. She produced her own gospel albums and hosted a radio show called The Dottie Peoples Showcase for six years.
Since joining Atlanta International Records in 1991, Dottie Peoples has released many successful albums, including live performances with a lively choir. Her music often appears on the Billboard Gospel chart, with her hit song "On Time God" (1994) staying in the top ten for two years. She has won several awards, including a Dove Award and multiple Stellar Awards, and has been nominated for Grammy and Soul Train awards. In 2003, she was inducted into the Gospel Music Hall of Fame.
The Talley Trio, a southern gospel group, has recorded Dottie Peoples's popular songs "He's an On Time God" and "Testify," reaching millions through concerts and TV performances. In 2008, she founded DP Muzik Group in Atlanta, producing a mix of traditional gospel and holy hip hop music.
Peoples has performed with Widespread Panic at the first Bonnaroo Music Festival and sung at significant events, including the 1996 Olympics and John Lewis's memorial service. She also holds an honorary Doctor of Sacred Music from the Global Evangelical Christian College and frequently tours with her friend Garnelle Hubbard-Spearman.
